چکیده

Acrylic type polymeric systems having degradable ester bonds linked to the 5-aminosalicylic acid (5-ASA), were synthesized and evaluated for colon targeted drug delivery. The obtained prodrug was characterized by FTIR,HNMR,Melting point and Rf value. In vitro drug release study was conducted at pH 1.2,7.4 and in rat fecal matter. Drug release in rat fecal matter at pH 7.4 was found to be most satisfactory. A burst release of 41.65% was observed in the first two hours followed by a sustained release over a period of 12 hours. A maximum of 92.46% of the drug was released from the prodrug and the time taken for 50% drug release (t50) was found to be nearly 3 hours. The best linearity for the prodrug was found in Higuchi’s equation, where r value was 0.9852, indicating the release of drug from prodrug as square root of time dependent process based on Fickian diffusion. The result suggest that these systems could be useful for preparation of a controlled release formulation of 5-aminosalicylic acid to target colon.

Synthesis of a novel PEGylated colon-specific azo-based 4- aminosalicylic acid prodrug

Objective(s): 4-aminosalicylic acid (4-ASA) is an isomer of mesalazine that has recently been shown to be effective against inflammatory bowel disease (IBD), and more specifically, ulcerative colitis. However, the majority of orally administered 4-ASA is readily and extensively absorbed from the stomach and small intestine, so only a small amount is transported to the ...

Synthesis and Pharmacological Evaluation of Cyclodextrin Conjugate Prodrug of Mefenamic acid

In the present investigation mefenamic acid prodrug of ß-cyclodextrins was synthesized. The primary hydroxy group of ß-cyclodextrins was used to block the acid group. The synthesis involved a series of protection and deprotection reaction. The ester was evaluated for stability in simulated gastric and intestinal fluid. The hydrolysis of cyclodextrin conjugate in colon is confirmed by the hydrol...

Corn Cob Xylan-based Nanoparticles: Ester Prodrug of 5-Aminosalicylic Acid for Possible Targeted Delivery of Drug

Xylan, a green biopolymer extracted from agro-waste corn cob may be used as a tool to deliver drugs principally to the colon because of their timely retention in the physiological environment of stomach and small intestine and can only be degraded in colon by vast anaerobic microflora such as Bifidobacterium. Present research work relates to incorporate the drug molecule such as 5-aminosalicyli...

Preparation of Ethylcellulose Coated Gelatin Microspheres as a Multiparticulate Colonic Delivery System for 5-Aminosalicilic Acid

In the long-term management of ulcerative colitis patients, repeat dosing maybe required. Since 5-ASA is largely absorbed from the upper intestine, selective delivery of drugs into the colon may be regarded as a better method of drug delivery with fewer side effects and a higher efficacy. The aim of this study was to prepare and evaluate a double coated multiparticulate system for 5-ASA deliver...
